Following phlebotomy, serum was separated and stored at — 20°C until analyzed. SACE activity was determined by a reference laboratory by measuring the release of the dipeptides benzoyl glycine and histidyl leucine from the synthetic substrate benzoyl-glycyl-histidyl-leucine. Lysozyme activity was determined locally by the lysoplate technique.
The subjects with sarcoidosis were included as positive controls for the measurement of SACE and lysozyme. The diagnosis of sarcoidosis was based on the presence of non-caseating granulomas found by tissue biopsy in the setting of typical clinical features, ie} lymphadenopathy, pulmonary involvement, cutaneous plaques, or laboratory findings, ie, hypergammaglobulinemia, hypercalcemia, or hypercalciuria.
The results suggest that underground coal mining, but not CWP, is associated with elevations in SACE and that after removal from the exposure to mixed coal mine dusts, the levels of SACE normalize.
